ASTM E2180 Standard Test Method for Determining the Activity of Incorporated Antimicrobial Agents in Polymeric of Hydrophobic Materials
Purpose of ASTM E2180 Test
ASTM E2180 test method is designed to quantitatively analyse the effectiveness of an antimicrobial agents incorporated into or onto a hydrophobic or polymeric surfaces. On hydrophobic surfaces, it is hard to spread the microorganisms. Proper microbial contact to an antimicrobial surface is necessary to evaluate the antimicrobial activity.
ASTM E2180 test method based on agar slurry inoculum facilitate good contact of microorganisms with antimicrobial treated hydrophobic surfaces. Therefore, it is great help in evaluating antimicrobial activity of hydrophobic antimicrobial treated surfaces.
ASTM E2180 Test Method
ASTM E2180 test method involves thin layer of the microorganisms inoculated agar slurry. This microbial inoculated agar slurry is then spreaded onto the test and untreated control material (triplicate samples minimum). After the specified contact time (24 h commonly used), surviving microorganisms are recovered via elution of the agar slurry inoculum from the test substrate into neutralizing broth. Serial dilutions are made, plated and incubated for 48 h at a specified temperature dependent upon the optimal temperature for test organism. Bacterial colonies from each dilution series are counted and recorded. Calculation of percent reduction of bacteria from treated versus untreated samples is made. Exposure periods may be modified (up to 96 h) to address more resistant microorganisms..

between untreated plastics or polymers and those with bound or incorporated low water-soluble antimicrobial agents.
ASTM E2180 test also help in determining the Comparisons between the numbers of survivors on preservative treated and control hydrophobic surfaces.
ASTM E2180 test procedure also permits determination of shelf-life or long term stability of an antimicrobial treatment which may be achieved through testing both non-washed and washed samples over a time span.
Turn Around Time: Eight to ten working days (if commonly tested organisms are tested.)
Sample Requirements for ASTM E2180 Test:
3.0 x 3.0 cm square samples of the treated and
control test materials (
Number of Samples: Since each time point requires 3 test pieces, for example for time 0 hour and 24hours= 3x2 = 6 pieces will be required per organism ordered.
An untreated control sample is also required.
Test Organisms:
Staphylococcus aureus ,Pseudomonas aeruginosa or Klebsiella pneumoniae .Other microorganisms such as yeast or fungi
may also be tested using this procedure.
